Being sued for DNA testing fees

Can my child's step-mother sue me for half of the costs of DNA testing if it comes back that her husband did not father my child?

The step mother has no standing to sue in the scenario that you relate. Certainly, if there is a paternity action pending, the court may require you to pay the costs of testing if the person tested turns out not to be the father.
