My tax preparer misled me into thinking I could get a tax credit that was improper and now I owe a large sum of money to IRS. Does their E&O insurance cover this or a portion of what I owe?

No! You have lost nothing with regard to the tax. If there is a penalty, you may be able to make a claim, but you would be far better getting a good tax lawyer to help you with the IRS and tax planning in the future.
