What are the characteristics required for reasonable suspicion to stop and briefly detain for questioning?

Just that--a reasonable suspicion. A reasonable suspicion exists somewhere between a hunch and probable cause. There is no standard test or set of circumstances that determines a reasonable suspicion and the existence, or not, of a reasonable suspicion, in a judicial setting, is determined separately on each set of facts or circumstances.
