My Doctor ordered some blood tests. The lab that performed the tests charged an astronomical price, 554.00 compared to 60.00 at another lab. I have called and talked with them about lowering the amount, to no avail. If I write a check PIF for a lesser amount and they cash it, is my responsibility done?

No, because there is no agreement to a lesser amount. You can send the provider a check for a lesser amount along with an offer to compromise the balance. If the check is then accepted, then you have a better claim of satisfaction and accord.
