Generally, no, but the one-word answer above leaves out a lot.

When an appeal is denied, the next step is to petition a higher court to review the decision.

There are certain situations where a decision can be appealed twice; for instance, denial of a motion to suppress evidence can be appealed prior to conviction and again after a conviction, as long as certain deadlines are met.
