Legal Question in Criminal Law in California

My brother is currently facing criminal charges and can no longer afford an attorney. Who and how do I ensure the court will appoint him a public defender? How do I follow up with this defender to ensure he is actively working on my brothers case?

Well you can't ensure an attorney is working on a case. Your brother needs to have his attorney request an appointment.

Usually the public defender won't "actively work" on the case. Your brother could possibly write to the public defender, by certified mail if possible, specifying which witnesses the public defender should interview and/or subpoena. This would create a record that could possibly help your brother in a future claim of ineffective assistance of counsel or legal malpractice.
