If you have 2 defendants and one answers the complaint and the other defendant demurrers to the complaint can the plaintiff amend the complaint without asking leave of the court

Yes. But most attorneys and trial court judges do not know it. See Barton v. Khan (2nd Dist. 2007) 157 Cal.App.4th 238.
