I will respond to your questions by number. 1. This question is overbroad. 2. Two is the minimum number needed to form a labor union. However a "Union" must secure the authorization of a majority of employees (either by written authorization or by a secret ballot election) to gain the right to represent employees as their colletive bargaining representative. 3. No. Supervisors and managers are excluded from collective bargaining units.
