Is it legal for a manager to deny her employees access to a company provided break room?

That is not a legal issue. As long as the employees are given their statutory break periods, an employer is under no obligation to provide a break room. Whether a manager is allowed to bar access to an existing break room is between the manager and the manager's superiors.
