Well my situation is that i work for a big chain retailer ive worked there for 10 years there i met my husband there and hes been there for 10 1/2 years. Weve been married for 6 years. At the store they never told us that one of us had to leave and hes s department manager but not above me. We are not the only couple that work at lowes also. But now corprate is wanting to make one of us transfer which is going to be me to snother store but i have to go thru the whole interview process. Can they do that

Yes, an employer can require a married couple to work in different locations. While it would be a better management practice to have a uniform policy followed throughout the company, there is nothing to prevent management from exercising its discretion to separate a couple, unless it is done for an unlawful reason, which has not been indicated in your post.
