Is booth rental considered subletting?

I run a spa in a commercial suite and would like to rent out one of the rooms to another licensed skincare professional. She will be operating out of my establishment using her own equipment and collecting her own dollars. I signed a lease in the beginning of the year with the commercial landlord. A section of the lease states:

"LESSEE shall not assign any portion of premises without prior written consent of the LESSOR, which shall not be unreasonably withheld, any such assignment of subletting without consent shall be void and, at the option of LESSOR, may terminate the lease. "

Would this time of arrangement require me to gain written consent from the Lessor?

Absolutely. What you want to do is exactly what that provision of the lease covers.
