Is an email confirmation of an agreement as binding as a written confirmation on letterhead with signatures?

Your question really goes to evidence. Unless there is a statute of frauds question, then an agreement is made when both sides mentally agree. As long as the contract is never litigated, then you have a binding agreement. As a litigator, I would prefer a signed contract with witnesses if I were to go to court to enforce the contract. But, it may still be enforceable.
You could follow up to the other side with a letter indicating that you have recieved an email confirmation and request that if it is in error, that the other side should respond in writing by a certain date. Send the letter certified and save the reciept.
