My friends husband- to-be last name and her mothers maiden name are the same, her husband will be petitioning for her in Florida, could she be asked to bring her mothers birth certificate to prove that her husband and mother are not related.

Yes. Birth certificates are always required. Note that a marriage is valid for USCIS purposes if it is valid in the jurisdiction where the marriage occurred. In some states, a marriage between first cousins is legal and would be honored by USCIS. Marriages between relatives of closer degree would never be recognized.
