Me and my wife moved to Florida 3 years ago from NY. My wife quit her job in NY in order to move to Florida. She has been searching for a job ever since. Isshe eligible to collect Unemployment Insurance?

Probably not, unless she was forced to quite because you were the primary breadwinner and she was following you, as she was the secondary breadwinner. It will depend on New York unemployment compensation law.
You are probably going to have a problem because of the time period in which she has between moving to Florida and leaving her job in New York.
Although New York law applies the Workforce Florida office in your area takes the application. Since it is an out-of-state application for benefits it must be done over the phone (maybe in person) and not on the computer as Florida employees are required to do.
