If you are disabled and cannot work and have a letter from Doctor. Do you have to deplete all you assets(savings, checking etc.), before you can file a claim for benefits?

No, social security diability is not based on indigency (poverty). As long as you are unable to be substantially, gainfully employed due to disability (which means your disability prevents you from making more than about $980 per month), you can get social security disability. They don't ask you about your assets.
