I am not married to my baby's dad and he offers no support of her. In the event of my death, what do I need to do to insure that he cannot have her or none of my assets (home, 401K etc.) I want her to be raised by my sister and her husband.

The father automatically gets custody assuming his paternity has been shown.
You can use a will to place assets in trust so that he can't benefit from assets for your child, and, since he may never step forward, you should also name a guardian in your will (although the father comes ahead of the guardian).

As to custody, the father, being the natural guardian, will get custody, unless he can be shown to be an unfit parent.
As to your assets, you can protect those by a properly worded will. Consult with a local attorney.
