My nephew has foced me to sign on a stamp paper that I have taken Rs.50Lakh from him as loan against my flat. How can he misuse it?

you need to see a lawyer and he would send file a civil suit restraining your nephew from relying upon the stamp paper and also seek cancellation of the paper.
