Can a father do DNA involving his daughter without the mothers consent? To find out if this child is his?

Legally yes, the test may be done on a child and father, however the result may not be as clear and convincing as it would be with the mother's cooperation.
If you and the mother have some genetic similarities then there may be some overlap in genetic markers. All of the child's markers will appear in either the father or the mother. Any deeper explaination would require a scientist, not a lawyer
