I am in USA with B/2 Visa, can I apply for E-2 visa

Yes, you can file a change-of-status to the E-2 prior to iration of the B-2 period of authorized stay, assuming that your country has an E-2 treaty with the US, and you qualify for the E-2 in all regards.
