Can a city or municipality be held accountable for a person being denied employment if that city or municipality was negligent in not removing a misdemeanor as they were supposed to?

No. Governmental immunity applies to ministerial acts.

No. Unlike employees in the private sector, government employees cannot be held accountable for neglect of duty. The legal system would be overwhelmed otherwise.
