Legal Question in Real Estate Law in Missouri
I bought a car in the state of MO. My friend helped me get a loan. He was the primary owner. I made the payments and used the car. I have repayed the loan now. I have the bank's letter. We have not received the car title from MO DMV/my friend lost it. I have applied to MO DMV thrice for change of ownership of the vehicle. My friend and I are not in good terms, he is not helping with this anymore. Car license plates are expired now. I urgently need to get my title to register it in the new state of residence. I want to take legal action against my so called friend, compelling him to do the necessary paper work to get me the title. What do I need to do?

It sounds like you need to get a Declaratory Judgment saying you own the car. That will allow you to get a new title in your name. You will probably need to hire counsel to bring this for you.
