Yes. Lender requires the signatures of all the people on the Deed title in order to get a good mortgage.

However, not all named owners on the Deed need to sign the Mortgage NOTE, which is the obligation to pay, if the loan was made based on the credit of fewer than all owners.

Whether this is related to a divorce, or the estate of a relative, or a business partnership which is breaking up, you need some legal advice. It may not be terribly complicated, and should not cost all that much money.
