What is the time limit on defamation of character in New York State? Does this apply to employee / employer relationships?

The statute of limitations for defamation is one year from the date of publication. It applies to all statements, verbal or written.

Whether an utterance constitutes slander or libel requires thorough analysis, especially in a situation where a "privilege" may attach to the statement such as in the workplace.
