Can employer ask an employee to sigh a letter of agreement not to file for unemployment if terminated at a later date?

Nope. They are violating Ohio law. Now, here comes the tricky part--do you want to confront your employer with this information and possibly get fired? Or do you have enough rapport with them to "educate" them. Don't know this answer. You will need to decide.
