I have an issue with a transfer on death deed. May I challenge the validity the deed same as I would a will with the defenses of undue influence, duress, etc? Further, where would I challenge the deed given that the deed avoids probate?

It's more difficult, but you can. It would be possibly be brought in common pleas general division court but may also be brought in probate court. This is one of those borderline jurisdiction matters.
