Calculation of 40 Hours for OT

If an employer pays and employee 2.5 times his or her hourly rate for such time worked on a holiday, can the company Not use those hours toward the calculation of 40 for overtime pay? or are they obligated to count all hours worked toward 40 irregardless of the rate of pay received for them?

The latter is true. They count actual hours worked, and not the dollar amount paid.
