In the state of Texas is it concidered statutory rape for a 21 year old male to have relations with a 16 year old female even if her parent is ok with it?

No. The affirmative defense only applies to sex with a 16 year old if you are less than 3 whole years older than her. You are at least 4 years older than her so you could still be prosecuted.
