It is possible to divorce a spouse that cannot be found. It is much easier if there are no children or property. Even though it is possible, the court will require the spouse seeking the divorce to make every effort to find that missing spouse. So he should start there: looking for that spouse. He should also document all efforts to locate the other spouse. Start by making a list of the other person's last known whereabouts, friends, family members, etc. Then get out the phone book and start calling people. Start searching the internet etc. If she is found, that will make things easier and less costly. If after extensive documented efforts fail, then he could seek a divorce and ask for "Service by Publication."
