Legal Question in Civil Rights Law in Virginia
While going through a security clearance, an old boss claimed I had a drug and alcohol problem. I left that prior job mostly due to her dumping all her work on me. I even wrote a letter to her boss stating the issues I was having and why I left. I was granted my security clearance and they found her allegations false. Do I have grounds to sue for defamation of character since this will forever show on my clearance record?

Yes, quite possibly if you have clear and convincing evidence that this "old boss"
not only communicated (published) false information that affected you in your career but at the time knew such to be false and without foundation.
I would recommend that you seek a consultation with a Virginia attorney
who is appropriately experienced and knowledgeable with respect
to how defamation claims are handled in the Commonwealth and who should
be able to advise you, accordingly, in regard to your situation.
