Cashier and money

My question is, does a cashier in a convenience store have the right to count their own till and if the owner will not permit this, are they responsible for any discrepancies?

No, unless the owner gives them such a right, and, yes, the owner could hold them responsible for such discrepancies if the owner so chooses, (however, if the owner falsely accuses a cashier, the owner could be civilly liable to this person so accused).
