I was hired part time 5 years ago. My new manager gave me a great review, then said I need you full time and if you don't work full time I will replace you. I felt threaten. Can she do this, If I'm under Dr's care for stress and depression.

Yes, assuming that you have no contract of employment which could prevent it.
