My wife was terminated from her job as a deli worker at a local grocery store. They claim that she ate food not paid for. They also claim they have her on video tape. My wife admitted to eating some food over the three year period they investigated. I just recieved a bill for $250 that they say is the amount she ate without paying for, is this leagal?

There is nothing illegal about it. If you contest the amount claimed, you may refuse to pay. The employer's remedy is to file a lawsuit, usually in Conciliation court, to seek payment. Chances of the employer pursuing such an action given the minimal amount in controversy seems unlikely.
