If I use a three wheel bicycle for a wheelchair, does a business have to let me enter and use the bike in their place of business?

Yes, but... Businesses are obligated to attempt reasonable accommodation of disability. That does not mean that you get to dictate to them what they must do. Your request must be about a provable and legitimately recognizable disability. Your request must be 'reasonable' under the circumstances. If your bike is too big for operation that is effective and safe for you AND other customers, then no, you'll have to use a real wheelchair.
