Can an USA citizen be brought back to Iran to be put on trial for rape and threatening to kill the person, since the crime took place in Iran?

Without an extradition treaty between the countries, no. But 'rendition' and 'termination with extreme prejudice' can be done privately, not just by CIA/NSA. It's called vigilante justice. I'd watch over my shoulder and not walk in any dark alleys if I were you.

And, if your response is 'but that's illegal', remember so is rape. Being illegal doesn't mean nobody does it. I'd watch over my shoulder and not walk in any dark alleys if I were you.

Since the US and Iran have not had diplomatic relations since 1976, I doubt there is an extradition treaty between the two countries. It is always possible that someone who is wanted for an alleged crime in Iran could be illegally kidnapped and taken there.
